Site Specific PEGylation

Currently available PEGylation technologies produce products which are PEGylated at multiple sites in an uncontrolled manner. This inevitably results in a population of molecules PEGylated in locations which crowd the active centre or binding regions resulting in a net decrease in potency versus the un-PEGylated analogue.

 The Almac unique approach enables homogeneous PEGylated populations where the specific PEGylation site can be selected and controlled, thus improving efficacy compared to existing PEGylation techniques, decreasing cost of goods and substance dosed to patient; while still providing an improved ADME profile due to the addition of the PEG moiety.

This PEGylation technique can be applied to all manner of peptides and proteins including cytokines, enzymes and antibody fragments.

Background Technology

Proprietary protein ligation technology

  • Site specific conjugation of synthetic molecules to the C-terminus of recombinant proteins
  • Labelled semi-synthetic proteins

Chemical synthesis & folding of small proteins

  • Up to 150-200 residues
  • Allows incorporation of labels or unnatural amino acids at any position
